摘要：A method of gene amplification which comprises annealing primers to a targetgene of an animal, a plant, etc. and then amplifying the gene by using a heat-resistantnucleic acid polymerase such as Taq polymerase or Pfu polymerase, characterized byusing at least one high-pressure fluid selected from the group consisting of supercriticalfluids such as carbon dioxide, subcritical fluids, high-pressure gases and liquids as thereaction medium. It is preferable to amplify the gene by using a surfactant-coatedenzyme obtained by coating a heat-resistant nucleic acid polymerase with a surfactantand controlling the reaction temperature.
